yojimbo can be fought numerous times, although the shortest path through the game has you not fighting him at all simply by skipping him more often than not people will fight him at least 3 time(first battle, he reapears but weaker, at the end of the game). if you use the save sphere or leave the area before getting him to join you will have to battle again.

my friend says 4 times
first is where you turn around and hes there.second is half way through but weaker.which doesnt really help coz hes got a massive amunt of health anyway.and the third is wehen your exiting the cave.

if you pop back to the save point he returns and you have to fight him once more, keep popping back to the save point and you can fight him more times than is even nesseary.
